The animal was perfused using Autoperfuser at a pressure of 130 mmHg with 300 ml 4% FA before being processed for paraffin embedding. HIER: Tris-EDTA, pH 9 for 20 min using Thermo PT Module. Blocking: 0.2% LFDM in TBST filtered thru 0.2 um. Detection was done using HRP polymer following manufacturers instructions; DAB chromogen: Candela DAB chromogen from Osenses. Primary antibody: dilution 1: 2000, incubated 30 min at RT using Autostainer. Sections were counterstained with Harris Hematoxylin.

Background: KCNJ10

FUNCTION: May be responsible for potassium buffering action of glial cells in the brain. Inward rectifier potassium channels are characterized by a greater tendency to allow potassium to flow into the cell rather than out of it. Their voltage dependence is regulated by the concentration of extracellular potassium; as external potassium is raised, the voltage range of the channel opening shifts to more positive voltages. The inward rectification is mainly due to the blockage of outward current by internal magnesium. Can be blocked by extracellular barium and cesium.; Subcellular location: Membrane, Multi-pass membrane protein.
