Western Blot: Kos1/Tnk1 Antibody [NB300-237] - T47D cells expressing GFP/GFP-Kos were lysed in RIPA lysis buffer. 500ug protein from clarified cell lysates was immunoprecipitated with an anti-GFP antibody. The washed immunoprecipitates were electrophoresed on 8% SDS-PAGE gel and the proteins were transferred to nitrocellulose membrane. Panel A indicates GFP and GFP-Kos when probed with anti-GFP antibody. The blot was stripped thereafter, and GFP-Kos was detected with Kos1/Tnk1 antibody (NB300-237) at a dilution of 1:1000

Background: Kos1/Tnk1

Kinase of embryonic stem cells (Kos1) is a nonreceptor protein tyrosine kinase (NRPTK) and a new member of the Ack family of NRPTKs. Kos1 is a ubiquitously expressed 47-kDa protein with autotyrosine kinase activity that is developmentally regulated during embryogenesis. Kos1 is also upregulated following IL3 withdrawal from factor-dependent murine NSF/N1.H7 cells that undergo apoptosis, suggesting a role in growth inhibition. Data support a negative regulatory role for Kos1 in regulating the Ras-Raf1-MAPK growth pathway by a mechanism that requires its autotyrosine kinase activity.
