Knockout Validated: Lactate Dehydrogenase B Antibody [NBP2-53421] - Western blot shows lysates of 293 human embryonic kidney parental cell line and LDHB knockout (KO) 293 cell line. PVDF membrane was probed with 1 ug/ml of Rabbit Anti-Human LDHB Polyclonal Antibody (NBP2-53421) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (HAF008). Specific band was detected for LDHB at approximately 35 kDa (as indicated) in the parental 293 cell line, but is not detectable in the knockout 293 cell line. This experiment was conducted under reducing conditions.

The membrane was probed with 1.0 ug/ml anti-LDHB in 1% non-fat milk in TBST and detected with an anti-rabbit HRP secondary antibody using chemiluminescence.

Immunocytochemistry/Immunofluorescence: Lactate Dehydrogenase B Antibody [NBP2-53421] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton X-100. The cells were incubated with anti-LDHB at 2 ug/ml overnight at 4C and detected with an anti-rabbit DyLight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Background: Lactate Dehydrogenase B/LDHB

LDHB, also known as L-lactate dehydrogenase B chain, is a member of the lactate dehydrogenase family. It is an oxidoreductase which catalyses the interconversion of pyruvate and lactate with concomitant interconversion of NADH and NAD+. As this protein can also catalyze the oxidation of hydroxybutyrate, it is occasionally called Hydroxybutyrate Dehydrogenase (HBD). The LDH family consists of three members, LDH-A, LDH-B and LDH-C. LDHs function as powerful markers for germ cell tumors. Recombinant human LDHB protein, fused to His-tagging at N-terminus, was expressed in E.coli and purified by using conventional chromatography techniques.
