Western Blot: MARCKS Antibody [NBP3-05540] - Western blot analysis of different cell lines lysates using MARCKS, at a dilution of 1:1,000 in green: [1] protein standard (red), [2] HEK293, [3] HeLa, [4] SH-SY5Y, and [5] COS1 cells. Strong band at 80kDa corresponds to MARCKS protein. The same blot was simultaneously probed with mouse mAb to GAPDH at a dilution of 1:5,000 in red, which reveals a single band at 37kDa in all preparations.

Immunocytochemistry/Immunofluorescence: MARCKS Antibody [NBP3-05540] - Immunofluorescent analysis of HeLa cells stained with MARCKS Antibody at a dilution of 1:5,000 in red, and costained with mouse mAb to beta-tubulin at a dilution of 1:10,000 in green. The blue is Hoechst staining of nuclear DNA. The MARCKS antibody binds to MARCKS protein expressed in the plasma membrane and cytoplasm, while the beta-tubulin antibody stains cytoplasmic microtubules.

Background: MARCKS

Myristolyated alanine rich C-kinase substrate, hence MARCKS, was originally discovered as a major substrate for protein kinase C in the brain and other tissues, and was originally isolated from human epithelial cells. MARCKS is a major protein of brain, and is concentrated in synapses of neurons. It appears to function in synaptic vesicle cycling and has been shown to bind both actin and calmodulin in vitro. MARCKS belongs to a family of proteins with similar actin and calmodulin binding properties. Deletion of the MARCKS gene in mice results in embryonic brain defects and death.
