Detection of Mouse CCR5 by Western Blot.
Western blot shows lysates of RAW 264.7 mouse monocyte/macrophage cell line treated (+) with 100 µg/mL LPS for 4 hours. PVDF Membrane was probed with 2 µg/mL of Rat Anti-Mouse CCR5 Monoclonal Antibody (Catalog # MAB6138) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). A specific band was detected for CCR5 at approximately 50 kDa (as indicated). Background: CCR5
CCR5 (CC chemokine receptor 5; also CD195 and MIP-1a receptor) is a 41 - 44 kDa member of the G-protein coupled receptor #1 family of proteins. It is expressed on Treg cells, NK cells, neurons, macrophages and Th1 cells. CCR5 mediates cell adhesion and migration induced by several chemokines including CCL3/MIP-1 alpha, CCL4/MIP-1 beta, CCL5/RANTES, and CCL8/MCP-2. It also functions as a coreceptor for macrophage-tropic HIV-1 infection. CCR5 contains an O-glycosylated and sulfated extracellular N-terminus (aa 1 - 32), and a phosphorylated and palmitoylated intracellular C-terminus (aa 304 - 354). CCR5 will form homodimers, heterodimers with CCR2, and heterooligomers with CCR2 and CXCR4. Within aa 1 - 32, mouse CCR5 shares 72% and 91% amino acid sequence identity with human and rat CCR5, respectively.
