Background: mu Opioid R/OPRM1

Opioid receptors are the membrane proteins that mediate the pain-relieving effect of opioid drugs, such as morphine and fentanyl as well as endogenous opioid peptides enkephalins and endorphins. The effects of opiates are mediated through opioid receptors located on the plasma membrane of neurons and other peripheral cells. There are at least 3 types of opioid receptors, mu, kappa and delta, each with a distinct pharmacologic profile (1). The endogenous opioid system is involved in stress responses, in the regulation of the experience of pain, and in the action of analgesic opiate drugs. The activation of the mu-opioid receptor system was associated with reductions in the sensory and affective ratings of the pain experience, with distinct neuroanatomical involvements. Data demonstrate the central role of the mu-opioid receptors and their endogenous ligands in the regulation of sensory and affective components of the pain experience (2).
