Immunocytochemistry/Immunofluorescence: Paxillin Antibody (LD4 sAB) [NBP2-81063] - Immunofluorescence analysis of paraformaldehyde fixed HeLa cells on Shi-fix coverslips, permeabilized with 0.15% Triton and stained with the chimeric mouse IgG1 version of LD4 sAB at 10 ug/ml for 1h followed by Alexa Fluor 488 secondary antibody (2 ug/ml), showing nuclear periphery and diffuse cytoplasmic staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP2-81063, DAPI, merged channels and an isotype control. Application Notes
This full-length, chimeric rabbit antibody was made using the variable domain sequences of the original Mouse scFv format, for improved compatibility with existing reagents, assays and techniques. This antibody binds to the LD4-motif of human paxillin, a signal transduction adaptor protein involved in focal adhesion, with binding partners including integrins, Src, FAK or actin complexes. The antibody inhibits complex formation between paxillin and FAK. Paxillin is involved in both inside-out cell signalling and outside-to-inside cell signalling.
