Immunoprecipitation:Proprotein Convertase 9/PCSK9 AntibodyNBP3-29941] -Detection of human PCSK9 by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from 293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-PCSK9 antibody NBP3-29941 (lot NBP3-29941-1) used for IP at 6 µg per reaction. For blotting immunoprecipitated PCSK9, NBP3-29941 was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 3 minutes.

Background: Proprotein Convertase 9/PCSK9

PCSK9 encodes a proprotein convertase belonging to the proteinase K subfamily of the secretory subtilase family. The encoded protein is synthesized as a soluble zymogen that undergoes autocatalytic intramolecular processing in the endoplasmic reticulum. The protein may function as a proprotein convertase. This protein plays a role in cholesterol homeostasis and may have a role in the differentiation of cortical neurons. Mutations in this gene have been associated with a third form of autosomal dominant familial hypercholesterolemia (HCHOLA3).
