Western Blot: PURA Antibody [NBP1-92651] - Samples: Whole cell lysate from HeLa (15 and 50 ug for WB; 1 mg for IP, 20% of IP loaded), 293T (T; 50 ug), Jurkat (J; 50 ug) and mouse NIH3T3 (M; 50 ug) cells. Antibodies: Affinity purified rabbit anti-Pur-alpha antibody NBP1-92651 used for WB at 0.1 ug/ml (A) and 1 ug/ml (B) and used for IP at 6 ug/mg lysate. Pur-alpha was more efficiently immunoprecipitated by rabbit anti-Pur-alpha antibody NBP1-92652, which recognizes a downstream epitope. Detection: Chemiluminescence with exposure times of 3 minutes (A) and 10 seconds (B).

Background: PURA

PURA product is a sequence-specific, single-stranded DNA-binding protein. It binds preferentially to the single strand of the purine-rich element termed PUR, which is present at origins of replication and in gene flanking regions in a variety of eukaryotes from yeasts through humans. Thus, it is implicated in the control of both DNA replication and transcription. Deletion of this gene has been associated with myelodysplastic syndrome and acute myelogenous leukemia.
