Background: Rad9

Rad9 is a recently isolated human protein that is involved in the control of cell cycle checkpoint. Human Rad9 (hRad9) is homologue of the S. pombe Rad9. Six Rad proteins (Rad1, Rad3, Rad9, Rad17, Rad26, and Hus1) are involved in the early steps of DNA damage check point response in S. pombe. Recently, it has been shown that hRad9 interacts with Bcl-2 family proteins Bcl-2 and Bcl-XL, but not with BAX or BAD. Over-expression of hRad9 in mammalian cells leads to apoptosis, where as antisense Rad9 suppresses cell death induced by DNA damaging agents such as methanosulphonate. Human Rad9 undergoes complex post-translational modifications in undamaged cells and is hyperphosphorylated in response to DNA damage and makes ternary complex with human Rad1 and human HUS1 proteins. This may account for detection of multiple protein bands in some cases by western blot or immunoprecipitation analysis using anti-hRad9 antibody. HRad9 has an intrinsic 3-5 exonuclease activity suggesting that by interaction with DNA it may play a role in the damage-sensing step of the DNA damage checkpoint pathway.
