Western Blot: RANK/TNFRSF11A Antibody (64C1385.1) [NBP2-24702] - Total protein from MG63, human testis and mouse testis was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-RANK1 in 1% block buffer and detected with an anti-mouse HRP secondary antibody using chemiluminescence.
Dual RNAscope ISH-IHC: RANK/TNFRSF11A Antibody (64C1385.1) [NBP2-24702] - Formalin-fixed paraffin-embedded tissue sections of human liver were probed for RANK/TNFRSF11A mRNA (ACD RNAScope Probe, catalog # 412281; Fast Red chromogen, ACD catalog # 322360). Adjacent tissue section was processed for immunohistochemistry using mouse anti-human (Novus Biologicals catalog # NBP2-24702) at 5 ug/mL with overnight incubation at 4 degrees Celsius followed by incubation with anti-mouse IgG VisUCyte HRP Polymer Antibody (Catalog # VC001) and DAB chromogen (yellow-brown). Tissue was counterstained with hematoxylin (blue). IHC signal is confined to plasma membrane.

Background: RANK/TNFRSF11A

Apoptosis, or programmed cell death, occurs during normal cellular differentiation and development of multicellular organisms. Apoptosis is induced by certain cytokines including TNF and Fas ligand of the TNF family through their death domain containing receptors, TNFR1 and Fas. Receptor activator of NF-kB (RANK) is a recently cloned member of the TNFR superfamily with no significant homology to other members of this family (1). RANK ligand (RANKL/TRANCE/ OPGL) (mAbs to RANKL is also available from Imgenex, Cat # IMG-133) binds to RANK on dendritic cells, upregulates the expression of anti-apoptotic protein BcL-XL suggesting a role in dendritic cell survival (2). The cytoplasmic domain of RANK interacts with TRAF2, TRAF5 and TRAF6 (3,4). Overexpression of RANK activates NF-kB and c-Jun-terminal kinase (JNK) pathways (3). Recent studies have shown that RANK interaction with TRAF6 activates NF-kB, whereas JNK activation is mediated through binding of RANK to TRAF2 (4).
