Recommended Usage

Neutralization

One neutralization unit is the amount of antiserum required to neutralize one unit of recombinant rat interferon beta (rat IFN-beta) to a 50% endpoint (one unit corresponds to approximately 15 pg of purified rat IFN-beta). Interferon was titrated with the use of the cytopathic effect inhibition assay as described [Rubinstein, S., Familletti, P.C., and Pestka, S. (1981) "Convenient Assay for Interferons," J. Virol. 37, 755-758; Familletti, P.C., Rubinstein, S., and Pestka, S. (1981) "A Convenient and Rapid Cytopathic Effect Inhibition Assay for Interferon," in Methods in Enzymology, Vol. 78 (S. Pestka, ed.), Academic Press, New York, 387-394]. In this antiviral assay for interferon about 1 unit/ml of interferon is the quantity necessary to produce a cytopathic effect of 50%. As there is no international rat IFN-beta standard,the units are determined with respect to the international reference standard for mouse interferon beta (mouse IFN-beta) provided by the National Institutes of Health [see Pestka, S. (1986) "Interferon Standards and General Abbreviations," in Methods in Enzymology (S. Pestka, ed.), Academic Press, New York 119, 14-23]. This material is prepared specifically for effective neutralization of rat IFN-beta.
Please Note: Optimal dilutions of this antibody should be experimentally determined.

Stability & Storage

Upon receipt, this product is stable at 4°C until the expiration date listed above. After reconstitution, the contents can be safely stored at 4°C for one month or at -20°C for one year for retention of full activity.When thawing, the contents of the tube should be apportioned in separate tubes so that freezing and thawing is kept to a minimum. Add 0.02% sodium azide to prevent bacterial growth.

Background: IFN-beta

Interferon-beta (IFN-beta) is a cytokine that is produced by fibroblasts and pathogen-exposed dendritic cells, macrophages, and endothelial cells. It signals through the heterodimeric IFN-alpha/beta Receptor. IFN-beta-deficient mice show increased susceptibility to experimental autoimmune encephalomyelitis (EAE), a disease model of human multiple sclerosis (MS). Furthermore, IFN-beta has been shown to suppress the Th17 cell response in both MS and EAE and has commonly been used as a treatment for MS.
