Recognizes the L1 or Calprotectin molecule (S100A9), an intra-cytoplasmic antigen comprising of a 12kDa alpha chain and a 14kDa beta chain expressed by granulocytes, monocytes and by tissue macrophages. This monoclonal antibody reacts with neutrophils, monocytes, macrophages, and squamous mucosal epithelia and has been shown as an important marker for identifying macrophages in tissue sections.

Flow Cytometry: S100A8/A9 Antibody (MRP8 7C12/4) - Azide and BSA Free [NBP2-47667] - Mouse splenocytes were fixed and permeabilized with Foxp3 Fixation/Permeabilization kit. Splenocytes were incubated with Fc block for 5 mins at RT and stained with or without antibody at 1:100 dilution for 25 mins at RT. Data was acquired on a Cytoflex LX and analyzed using FlowJo 10. Application Notes
Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.
Optimal dilution for a specific application should be determined.
Please Note: Optimal dilutions of this antibody should be experimentally determined.

Background: S100A9

S100A9 is a member of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and are involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. S100A9 may function in the inhibition of casein kinase, and altered expression is associated with the disease cystic fibrosis. Overexpression of S100A9 has also recently been linked to a poor prognosis with invasive ductal carcinoma of the breast.
