Flow Cytometry: TGF-alpha Antibody (tAb2) - Chimeric - Azide and BSA Free [NBP2-81082]

Flow Cytometry: TGF-alpha Antibody (tAb2) - Chimeric - Azide and BSA Free [NBP2-81082]

Flow Cytometry: TGF-alpha Antibody (tAb2) [NBP2-81082] - Flow-cytometry using the [NBP2-81082]. Human peripheral blood leukocytes were fixed using 2% PFA, permeabilised using 0.5% Triton and stained with unimmunized rabbit IgG antibody (isotype control, black line) or the rabbit IgG-chimeric version of tAb2 (NBP2-81082, blue line) at a dilution of 1:100 for 1h at RT. After washing, bound antibody was detected using a goat anti-rabbit IgG AlexaFluor 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

Applications for TGF-alpha Antibody (tAb2) - Chimeric - Azide and BSA Free

Application
Recommended Usage

Dot Blot

Optimal dilutions of this antibody should be experimentally determined.

ELISA

Optimal dilutions of this antibody should be experimentally determined.

Flow Cytometry

Optimal dilutions of this antibody should be experimentally determined.
Application Notes
This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgG1 format, for improved compatibility with existing reagents, assays and techniques. Human transforming growth factora (TGFa) consists of 50 amino acid residues with three disulphide bridges. It belongs to the family of epidermal growth factor related peptides and binds to EGFreceptor. It is involved in cell proliferation in healthy cells and is found in various cancer cells. Transfection of Fibroblasts with the TGFa gene induces tumor formation.

Background: TGF-alpha

Transforming growth factor alpha (TGFalpha) is an acid- and heat-stable 50 amino acid protein originally found in rodents and humans. TGFalpha is 33% homologous at the amino acid level to epidermal growth factor (EGF). TGFalpha binds to the EGF receptor, mediates tyrosine phosphorylation of the receptor and promotes anchorage-independent growth of normal rat fibroblasts in soft agar in the presence of transforming growth factor beta. TGFalpha is secreted by a variety of transformed cells and tumors, embryonic cells and some normal adult cells. TGFalpha bioactivity has been found in the urine of cancer patients. It has been suggested that it may act as an autocrine growth factor for the induction or maintenance of malignancy.
