Western Blot: TRA-1-60 (R) Antibody (TRA-1-60(R)) [NB100-730] - Whole cell protein from human ES cells (lane 1), human NTERA-2 cells (lane 2), mouse embryonic fibroblasts (MEF, lane 3) and mouse V6.5 ES cells (lane 4, NBP1-97433) were separated on a 7.5% gel and transferred to PVDF membrane. The membrane was then incubated with anti-TRA1-60(R) antibody at 2 ug/ml and detected with an anti-mouse IgM secondary antibody using chemiluminescence. Note how this antibody specifically detects the TRA1-60(R) epitope only on human cells.

Cells were stained using an Anti-Mouse IgG Secondary Antibody (red) and counterstained with DAPI (blue). Specific staining was localized to cell surfaces.

Background: TRA-1-60(R)

TRA-1-60 is a cell-surface protein involved in cell differentiation. Cell surface antigens are powerful tools for characterizing the function and expression of developmentally regulated cells during both embryogenesis and oncogenesis. The TRA-1-60 epitope is also found on human embryonic stem (ES) cells and primordial germ cells, and TRA-1-60 serves as a serum marker in patients with germ cell tumors. Investigation into the identity of the TRA-1-60 epitope demonstrated that it is a carbohydrate carried by a cell surface, sialylated, keratan sulfate proteoglycan. Subsequent evidence implicated podocalyxin as a carrier for the TRA-1-60 epitope.
