TRPV6 Antibody

Western Blot: TRPV6 Antibody [NBP3-33063]

Western Blot: TRPV6 Antibody [NBP3-33063] - Western blot analysis of TRPV6 on different lysates with Rabbit anti-TRPV6 antibody (NBP3-33063) at 1/500 dilution.

Lane 1: SW480 cell lysate
Lane 2: SK-Br-3 cell lysate

Lysates/proteins at 10 ug/Lane.

Predicted band size: 87 kDa
Observed band size: 42 kDa (unglycosylated monomeric form of TRPV6)

Exposure time: 30 seconds;

8% SDS-PAGE gel.

Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-33063) at 1/500 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:300,000 dilution was used for 1 hour at room temperature.
TRPV6 Antibody

Immunohistochemistry: TRPV6 Antibody [NBP3-33063]

Immunohistochemistry: TRPV6 Antibody [NBP3-33063] - Immunohistochemical analysis of paraffin-embedded rat testis tissue using anti-TRPV6 antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 8.0-8.4) for 20 minutes.The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-33063, 1/50) for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
TRPV6 Antibody

Immunocytochemistry/ Immunofluorescence: TRPV6 Antibody [NBP3-33063]

Immunocytochemistry/ Immunofluorescence: TRPV6 Antibody [NBP3-33063] - ICC staining of TRPV6 in HepG2 cells (green). Formalin fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 10 minutes at room temperature and blocked with 1% Blocker BSA for 15 minutes at room temperature. Cells were probed with the primary antibody (NBP3-33063, 1/100) for 1 hour at room temperature, washed with PBS. Alexa Fluor®488 Goat anti-Rabbit IgG was used as the secondary antibody at 1/100 dilution. The nuclear counter stain is DAPI (blue).

Background: TRPV6

Calcium-permeable channels, such as TRPV6, participate in neurotransmission, muscle contraction, and exocytosis by providing calcium as an intracellular second messenger. Depending on the tissue, transcellular calcium transport may be regulated by vitamin D, parathyroid hormone (PTH), or calcitonin (CALCA); FUNCTION: Calcium selective cation channel probably involved in Ca(2+) uptake in various tissues, including Ca(2+) reabsorption in intestine. The channel is activated by low internal calcium level, probably including intracellular calcium store depletion, and the current exhibits an inward rectification. Inactivation includes both, a rapid Ca(2+)-dependent and a slower Ca(2+)-calmodulin-dependent mechanism, the latter may be regulated by phosphorylation. In vitro, is slowly inhibited by Mg(2+) in a voltage-independent manner. Heteromeric assembly with TRPV5 seems to modify channel properties. TRPV5-TRPV6 heteromultimeric concatemers exhibit voltage-dependent gating. SUBUNIT: Homotetramer and probably heterotetramer with TRPV5. Interacts with TRPV5. Interacts with S100A10 and probably with the ANAX2-S100A10 heterotetramer. The interaction with S100A10 is required for the trafficking to the plasma membrane. Interacts with BSPRY. Interacts with calmodulin. SUBCELLULAR LOCATION: Cell membrane; Multi-pass membrane protein.
