Application Notes
This UBCH6 antibody is useful for Immunocytochemistry/Immunofluorescence and IHC-paraffin embedded sections. In ICC/IF, nuclear staining was observed in HeLa cells. In IHC-P, staining was observed in the nucleus and plasma membrane of mouse intestine tissue. Prior to immunostaining paraffin tissues, antigen retrieval with sodium citrate buffer (pH 6.0) is recommended. This antibody is not applicable for Western Blot.

Background: UbcH6/UBE2E1

Ubiquitination is a cellular process by which the cell identifies abnormal or short-lived proteins for degradation. Three classes of enzymes are involved in this process, ubiquitin-activating enzymes, or E1s, ubiquitin-conjugating enzymes, or E2s, and ubiquitin-protein ligases, or E3s. UBCH6 (UBE2E1 or ubiquitin-conjugating enzyme E2E 1) belongs to the E2 class of enzymes. ISG15, an ubiquitin like protein has been shown to covalently modify UBCH6 as well as forming a thoiester intermediate with ubiquitin. ISG15-modified UBCH6 formed a ubiquitin thioester while the unmodified UBCH6 did not which suggests that ISG156 conjugation suppresses UBCH6 E2 enzyme activity. (PMID: 16428300) In Spinalcerebellar Ataxia-1 (SCA1), research has shown that UBCH6 ubiquinates Ataxin-1. The overexpression of UBCH6 also reduced the transcriptional repression activity of ataxin-1. It addition it was found that overexpression of UbcH6 reduced the formation of Ataxin-1 aggregated. By modulating the degradation of ataxin-1, UbcH6 can regulate the transcriptional repression activity of ataxin-1 (PMID: 18519031).
