Immunogen

Antibody was raised against a peptide corresponding to 17 amino acids near the amino terminus of human XBP1. The immunogen is located within amino acids 40 - 90 of XBP1. Amino Acid Sequence: GLPQARKRQRLTHLSPE

Specificity

Human XBP1 has two isoforms, including isoform 1 (261aa, 29kD) and isoform 2 (376aa, 40kD). Mouse XBP1 also has two isoforms, including isoform 1 (267aa, 29.6kD) and isoform 2 (371aa, 40kD). And Rat XBP1 has two isoforms as well, including isoform 1 (267aa, 30kD) and isoform 2 (371aa, 40kD). NBP1-77253 can detect human, mouse and rat.

Description

CoraFluor(TM) 1 is a high performance terbium-based TR-FRET (Time-Resolved Fluorescence Resonance Energy Transfer) or TRF (Time-Resolved Fluorescence) donor for high throughput assay development. CoraFluor(IM) 1 absorbs UV light at approximately 340 nm, and emits at approximately 490 nm, 545 nm, 585 nm and 620 nm. It is compatible with common acceptor dyes that absorb at the emission wavelengths of CoraFluor(TM) 1. CoraFluor(TM) 1 can be used for the development of robust and scalable TR-FRET binding assays such as target engagement, ternary complex, protein-protein interaction and protein quantification assays.

Background: XBP1

XBP1 encodes a transcription factor that regulates MHC class II genes by binding to a promoter element referred to as an X box. The protein contains two domains commonly found in transcription factors that confer DNA binding and dimerization capability. This gene product was also separately identified as a cellular transcription factor that binds to an enhancer in the promoter of the T cell leukemia virus type 1 promoter. It may increase expression of viral proteins by acting as the DNA binding partner of a viral transactivator. There is also a recent finding that the same agents that cause an ER stress response, the unfolded protein response, also increase transcription of this gene. This increase in transcription requires an ER stress response consensus binding element in the promoter.
