Application Notes

1. Dilute the desired amount of Coating Antibody (COAT) (1 mg/ml) (rabbit polyclonal antibody) to 5 ug/ml in PBS without carrier protein and use it fresh.
2. Dilute the desired amount of Detection Antibody (DET) (0.5 mg/ml) (mouse monoclonal antibody) to 0.5 ug/ml in ELISA Buffer and use it fresh.
3. Reconstitute the Streptavidin-HRP (STREP) with 100 ul ELISA Buffer. After reconstitution, prepare aliquots and store the reconstituted STREP at -20C. Avoid freeze/thaw cycles. Dilute the reconstituted Streptavidin-HRP to the working concentration by adding 10 ul in 10 ml of ELISA Buffer (1:1000).
4. Reconstitute the Standard Protein (STD) (recombinant human IL-36 beta/IL-1F8) with 100 ul ELISA Buffer to obtain a concentration of 1 ug/ml. After reconstitution, prepare aliquots and store the reconstituted standard at -20C. Avoid freeze/ thaw cycles. A standard curve using 2-fold serial dilutions in ELISA Buffer is recommended. Suggested standard points are 1 ng/ml, 0.5 ng/ml, 0.25 ng/ml, 0.125 ng/ml, 0.062 ng/ml, 0.031 ng/ml, 0.0156 ng /ml and 0 ng/ml.

Background: IL-36 beta/IL-1F8

Interleukin 36 beta (IL-36 beta), also known as IL-1F8, FIL-1h and IL-1H2, is a member of the IL-1 family of proteins. IL-36 beta is reported to be actively secreted. Cells expressing IL-36 beta include resting and activated monocytes, and B cells. The receptor for IL-36 beta is thought to be a combination of IL-1 Rrp2 and IL-1 RAcP. Recombinant IL-36 beta activates pathways involving NF-kB and MAPK in an IL-1 Rrp2-dependent manner.
