The goal is to change the volume of the blood as little as possible when adding the indomethacin. We recommend the following procedure:1. Prepare a 10 mg/mL stock solution of indomethacin in ethanol. This can be stored in the freezer. (It will precipitate during storage, but can be redissolved once warmed to room temperature.)2. The day that samples will be drawn, dilute the indomethacin 1:50 in PBS for a concentration of 0.2 mg/mL.3. Immediately after the sample is drawn, add 50 uL of indomethacin (at 0.2 mg/mL) for every 1 mL of whole blood directly into the tube and mix gently.

Background: Thromboxane B2

Thromboxane A2 (TXA2), is an eicosanoid involved in platelet aggregation, vasoconstriction, and reproductive functions. However, as TXA2 is rapidly degraded in vivo, TXA2 production is typically examined by assaying the more stable Thromboxane B2 (TXB2) or 11-dehydro-TXB2 (11-d-TXB2) derivatives. Levels of these metabolites are helpful markers in the study of diseases including liver cirrhosis, cystic fibrosis, mastocytosis, systemic lupus erythematosus, thrombosis diseases, and others.
