Product Summary for DAPI Solution
DAPI is excluded by viable cells but can penetrate cell membranes of dying or dead cells, in which it intercalates into double stranded nucleic acids. Dead cells which take up DAPI will fluoresce brightly around 461nm. DAPI may be excited by the UV or violet lasers although the UV laser is much more efficient. DAPI's emission spectra is very similar to the Pacific Blue (TM) dye.

Immunocytochemistry/Immunofluorescence: DAPI Solution [NBP2-31156] - Staining image showing the use of DAPI solution (Blue) [NBP2-31156] as a counterstaining agent in ICC/IF analysis of 58K Golgi Protein and alpha tubulin in Hela cell line. Cells, cultured on cover slips, were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton-X100. The cells were incubated with 1:200 dilution of anti-58K Golgi Protein antibody clone 58K-9 for overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) secondary at a 1:500 dilution. Alpha tubulin (DM1A) [NB100-690] was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse Dylight 550 (Red) at a 1:500 dilution.
