Background: CD4

CD4 is a type I membrane glycoprotein belonging to the immunoglobulin superfamily. It is expressed predominantly on thymocytes and a subset of mature T lymphocytes (1). CD4 is a standard phenotype marker for the identification of T cell populations (1). CD4 functions in collaboration with the T cell receptor in the recognition of peptide antigens that are presented by class II major histocompatibility complexes (2, 3). CD4 also has been shown to be a coreceptor of HIV entry and specifically binds gp120, the external envelope glycoprotein of HIV (4). Mature canine CD4 consists of a 377 aa extracellular region containing four immunoglobulin-like domains, a 22 aa transmembrane segment, and a 40 aa cytoplasmic domain (5). Within the extracellular region, canine CD4 shares 57%, 46%, and 42% amino acid sequence identity with human, mouse, and rat CD4, respectively.
