Recombinant Human Meteorin-like/METRNL Induces Stat3 Phosphorylation in E16-E18 Rat Cortical Neurons. Treatments of E16-E18 rat cortical neurons with 1 μg/ml (1) and 5 μg/ml of Recombinant Human Meteorin-like/METRNL (2) (Catalog # 9339-MN) for 15 mintues results in Stat3 phosphorlation. Background: Meteorin-like/METRNL

Meteorin-like protein (METRNL, Subfatin, Cometin and Interleukin 39) is a 311 amino acid (aa) protein that shows 58% similarity with Meteorin (in human). Human Meteorin‑like protein is synthesized with a 45 amino acid (aa) signal peptide with the secreted mature protein ~30 kDa (1). Mature human Meteorin‑like protein shares 79% and 80% aa sequence identity with mouse and rat METRNL, respectively. Alternate splicing generates an additional isoform that lacks the N‑terminal 82 amino acids including the signal peptide (1). Metrnl is expressed in various adult mouse tissues with the highest expression in muscle tissue, adipose tissue, and activated monocytes (2, 3). Its expression is induced in muscle after exercise and in adipose tissue upon cold exposure suggesting that this secreted protein is both a myokine and an adipokine (4). Neurotrophic activity has been ascribed to Metrnl as it enhances neurite outgrowth and neuroblast migration in vivo. This effect is mediated by the Jak-STAT3 and MEK-ERK pathways (5). Metrnl has also been shown to be indirectly involved in the "browning" of white fat to a more thermogenic "beige fat" through activation of the STAT6 pathway in adipose tissue macrophages, inducing the M2-regulatory cytokines IL-4 and IL-13 (6). Metrnl transgenic mice have demonstrated a role for Metrnl in insulin sensitization through up-regulation of PPAR gamma, a key transcription factor regulating adipocyte differentiation. PPAR gamma increases adipocyte differentiation, inhibits adipose inflammation, activates lipid metabolism, and sensitizes adipocytes to insulin (4).
