Background: Plexin A3

Plexin A3, also known as Plexin-4, is a member of the large Plexin family of conserved transmembrane receptors that mediate Semaphorin functions (1, 2). There are four classes plexins (A, B, C, and D) and they share a similar domain structure: An extracellular domain (ECD) that binds the semaphorin ligand, a single transmembrane domain and an approximately 600 residue-long cytoplasmic region (2). The ECD of mature, human plexinA-3 consists of a Sema domain and four IPT (Ig-like, plexins, transcription factors) domains and shares 93% amino acid identity with both mouse and rat PlexinA-3. Plexin A3 is a coreceptor for class 3 semaphorins and plays an important role in axon guidance in the developing nervous system and regulates the migration of sympathetic neurons (3, 4). Class 3 semaphorins bind to a complex composed of a neuropilin and a plexin (4). Plexin A3 is required for normal dendrite spine morphology in pyramidal neurons and may play a role in regulating semaphorin-mediated programmed cell death in the developing nervous system (5). The cytoplasmic domain of plexinA-3 is required for the activation of down-stream signaling events in the cytoplasm (6).
