Background: SLITRK3

SLITRK3 (Slit and Trk-like family member 3) is an approximately 120 kDa type I transmembrane member of the SLITRK family of proteins which contain a Slit-like extracellular region and (save for SLITRK1) a Trk-like cytoplasmic region (1). The extracellular domain (ECD) of mature human SLITRK3 contains 6 leucine rich repeats (LRR) followed by a C-terminal LRR domain, followed by 6 more LRR flanked by a pair of N- and C-terminal LRR domains (2, 3). Within the ECD, human SLITRK3 shares 98% amino acid sequence identity with mouse and rat SLITRK3. It is expressed in multiple regions of the brain, particularly the cerebral cortex and hippocampus (4). SLITRK3 can suppress neurite outgrowth and promote the formation of inhibitory presynaptic structures (3-5). It localizes post-synaptically and binds to PTP-delta and PTP-sigma on pre-synaptic inhibitory terminals (4, 5). SLITRK3 is upregulated during the development of gastrointestinal stromal tumors (6).
