Background: Reg2
Mouse Reg2 (Islets of Langerhans regenerating protein 2), also known as Reg‑II, Lithostathine 2, PTP2 (pancreatic thread protein 2) and PSP2 (pancreatic stone protein 2), is a member of the Reg family of secreted C‑type lectin domain‑containing pancreatic proteins (1, 2). Reg2 forms a subfamily with mouse and human Reg1 molecules (1). The mouse Reg2 cDNA encodes 173 amino acids (aa) including the 22 aa signal sequence and the 151 aa secreted mature protein. Mouse Reg2 shares approximately 83% aa sequence identity with hamster Reg2, but no Reg2 ortholog exists in humans or rats (1, 2). Reg proteins are thought to be stress response proteins that stimulate proliferation, particularly in beta cells of pancreatic islets (1). Reg2 is expressed primarily by acinar cells in the normal exocrine pancreas, and is up‑regulated in NOD (non-obese diabetic) islets, hyperplastic islets, regenerating islets or in response to pancreatic injury or pancreatitis (1‑5). IFN‑ beta causes Reg2 up‑regulation in early‑onset diabetic NOD mice or in a NOD insulinoma cell line (4). Reg2 is also up‑regulated in mice lacking the pancreas‑expressed keratin K8, and is thought to be regulated by keratin filament organization (3). It is proposed to serve as an autoantigen in NOD mouse immune‑mediated type I diabetes models (4).
