ChIP Video Protocol
(These may take a moment to load. It will be worth the wait!)
Phase 1: Lysate Preparation and Chromatin Cross-Linking
In this video you will learn how to crosslink protein to DNA and prepare cells for chromatin shearing. Components of the Bio-Techne ChromataChIP kit, an all in one ChIP solution, are also shown. Full length protocols and additional help can be found in the support section, through our live chat service, or by calling us directly to talk with our elite customer and technical service scientists.
Phase 2: DNA Shearing by Sonication
In this video you will learn how to effectively shear chromatin for ChIP using a sonicator, and how to analyze DNA fragment sizes with an agarose gel. Components of the Bio-Techne ChromataChIP kit, an all in one ChIP solution, are also shown. Full length protocols and additional help can be found in the support section, through our live chat service, or by calling us directly to talk with our elite customer and technical service scientists.
Phase 3: Immunoprecipitation
In this video you will learn how to precipitate your DNA-protein complex of interest using an antibody and magnetic bead based system. Beads can be washed manually, or multiplexed processed with the aid of the Precipitor IP machine. Components of the Bio-Techne ChromataChIP kit, an all in one ChIP solution, are also shown. Full length protocols and additional help can be found in the support section, through our live chat service, or by calling us directly to talk with our elite customer and technical service scientists.
Phase 4: Reverse Cross-Linking and DNA Purification
In this video you will learn how to reverse the DNA-protein cross-linking, followed by DNA purification through silica based cleanup columns. Your precipitated and cleaned DNA can then be analyzed by a variety of detection methods discussed in the final phase. Components of the Bio-Techne ChromataChIP kit, an all in one ChIP solution, are also shown. Full length protocols and additional help can be found in the support section, through our live chat service, or by calling us directly to talk with our elite customer and technical service scientists.
Phase 5: DNA Amplification and Quantitation
In this video you will learn how load your precipitated and cleaned DNA into a microwell plate for amplification and detection through quantitative real-time PCR. Components of the ChromataChIP kit, an all in one ChIP solution, are also shown. Full length protocols and additional help can be found in the support section, through our live chat service, or by calling us directly to talk with our elite customer and technical service scientists.
