Background: ATOX1

ATOX1 (anti-oxidant 1 or HAH1) is an ATX1 family member and is a ubiquitously expressed metallochaperone which binds/deliver cytosolic copper to copper ATPase proteins. Copper is an essential trace element which at high concentration is extremely toxic and a balance between copper import/export is highly critical for survival. To ensure that the total pool of free copper is less than 1 ion per cell, ATOX1 specifically deliver copper to cellular compartments and copper-dependent enzymes. ATOX1 interacts with ATP7A (Menkes protein) and ATP7B (Wilson protein) in a copper-dependent manner leading to facilitation of copper transfer from ATOX1 to ATP7A or ATP7B, thus permitting copper transport into the secretory pathway and copper export from the cell. The conserved metal binding site of ATOX1, CxGC, differs from the metal-binding sites of Cu-transporting ATPases and has a physiologically relevant redox potential that equilibrates with the GSH: GSSH pair. Cellular functions of ATOX1 are not limited to its copper-trafficking role and may include storage of labile copper, modulation of transcription, and antioxidant defense.
