Western Blot: BACE-2 Antibody (1/ 9) [NBP2-81075] - Western Blot using anti-BACE2 antibody (1/9) [NBP2-81075]. HeLa (A) and U251 (B) cell lysate samples (35ug protein in RIPA buffer) were resolved on a 10% SDS PAGE gel and blots probed with the chimeric mouse IgG1 version of 1/9 [NBP2-81075] at 1 ug/ml before detection using an anti-mouse secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence. The predicted running size for BACE2 is 56.2 kDa. NBP2-81075 successfully detected BACE2 in HeLa and U251 cell lysate samples.

Immunocytochemistry/Immunofluorescence: BACE-2 Antibody (1/ 9) [NBP2-81075] - Immunofluorescence staining of fixed U251 cells with BACE-2 Antibody (1/ 9) [NBP2-81075]. Immunofluorescence analysis of paraformaldehyde fixed U251 cells permeabilized with 0.15% Triton and stained with the chimeric mouse IgG1 version of 1/9 [NBP2-81075] at 10 ug/ml for 1h followed by Alexa Fluor 488 secondary antibody (2 ug/ml), showing cytoplasmic staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP2-81075, DAPI, merged channels and an isotype control. The isotype control was stained with an anti-unknown specificity antibody followed by Alexa Fluor 488 secondary antibody.

Application Notes
This reformatted mouse antibody was made using the variable domain sequences of the original Mouse IgG format, for improved compatibility with existing reagents, assays and techniques. The antibody binds specifically to BACE2, an enzyme responsible for the proteolytic cleavage of amyloid precursor protein (APP). The protein is cleaved between residues 690 and 691, leading to the formation of beta-cleaved soluble APP and a C-terminal membrane bound fragment (CTFbeta). The latter is then cleaved by gamma-secretase to generate amyloid beta peptide. BACE is the rate-limiting enzyme in the pathway, and is a potential target in the treatment of Alzheimer's Disease (AD),a neurodegenerative disorder characterised by amyloid beta plaques within the brain. The BACE2 protein is encoded in chromosome 21, in a region critical for the development of Down's Syndrome, and may play a role in the amyloid beta deposition in the disease.

Background: BACE-2

BACE2 (Memapsin 1) is a membrane-bound aspartic protease that is highly homologous with BACE1 (Memapsin 2). While BACE1 processes the amyloid precursor protein (APP) at a key step in generating the beta-amyloid peptide and presumably causes Alzheimer's disease (AD), BACE2 has not been demonstrated to be directly involved in APP processing, and its physiological functions remain to be determined. In vivo, BACE2 is expressed as a precursor protein containing pre-, pro-, protease, transmembrane, and cytosolic domains/peptides.
