Product Summary for CDC2/CDK1 Antibody (CDK1/873) [CoraFluor™ 1]

Immunogen

Recombinant human full-length CDC2/CDK1 protein. (Uniprot: P06493)

Reactivity Notes

Does not react with Mouse, Rat, Xenopus, drosophila, S, pombe or S. cerevisiae.

Localization

Nuclear and Cytoplasmic

Specificity

Recognizes a 34kDa protein, which is identified as cyclin dependent kinase 1 (cdk1) or p34cdc2 protein kinase. cdk1 / p34cdc2 plays a crucial role during cell division and is most active during mitosis. It is predominantly localized in the nucleus. It is a serine/threonine kinase, which is activated by cyclin, presumably by de-phosphorylation of tyrosine residues. Activated cdk1 / p34cdc2 performs specific functions during mitosis, including nuclear envelope breakdown and chromosome condensation.

Clonality

Monoclonal

Host

Mouse

Isotype

IgG2a Kappa

Description

CoraFluor(TM) 1 is a high performance terbium-based TR-FRET (Time-Resolved Fluorescence Resonance Energy Transfer) or TRF (Time-Resolved Fluorescence) donor for high throughput assay development. CoraFluor(IM) 1 absorbs UV light at approximately 340 nm, and emits at approximately 490 nm, 545 nm, 585 nm and 620 nm. It is compatible with common acceptor dyes that absorb at the emission wavelengths of CoraFluor(TM) 1. CoraFluor(TM) 1 can be used for the development of robust and scalable TR-FRET binding assays such as target engagement, ternary complex, protein-protein interaction and protein quantification assays.

Background: CDC2/CDK1

The cell division control protein cdc2, also known as cyclin-dependent kinase 1 (Cdk1) or p34/cdk1, plays a key role in the control of the eukaryotic cell cycle, where it is required for entry into S-phase and mitosis. Cdc2 exists as a complex with both cyclin A and cyclin B. The best characterized of these associations is the Cdc2 p34 cyclin B complex, which is required for the G2 to M phase transition. Activation of Cdc2 is controlled at several steps including cyclin binding and phosphorylation of threonine 161. However, the critical regulatory step in activating cdc2 during progression into mitosis appears to be dephosphorylation of Tyr15 and Tyr14. Phosphorylation at Tyr15 and inhibition of Cdc2 is carried out by WEE1 and MIK protein kinases while Tyr15 dephosphorylation and activation of Cdc2 is carried out by the cdc25 phosphatase. The isoform CDC2deltaT is found in breast cancer tissues. Furthermore, cdc2/Cdk1 is a key mediator of neuronal cell death in brain development and degeneration.
