Specificity

Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. Most pituitary adenomas and prolactinomas readily express chromogranin.

Application Notes
Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.
Optimal dilution for a specific application should be determined.
Please Note: Optimal dilutions of this antibody should be experimentally determined.

Background: Chromogranin A

Chromogranin A (CgA) is an 86 kDa protein that is the major member of the granin family of acidic secretory glycoproteins located in neuroendocrine cells. CgA is believed to play a role in targeting peptide hormones and neurotransmitters to granules of the regulated pathways and inhibit hormone and neurotransmitter release. Also, the widespread distribution of CgA has made the measurement of circulating immunoreactive CgA a valuable tool in the diagnosis of neuroendocrine neoplasia, and CgA immunohistochemistry can help to identify the neuroendocrine nature of tumors. The N-terminal domain of CgA inhibits tumor necrosis factor a (TNFa) induced gap formation in human umbilical venous endothelial cells. CgA levels which reflect neuroendocrine differentiation of prostatic carcinoma may have a diagnostic, therapeutic and prognostic role in the management of prostate cancer patients.
