Western Blot: Clusterin/APOJ Antibody (SAIC-43B-8) - Azide and BSA Free [NBP3-20109] - HeLa(A) (0.03ug/ml), MCF7(B) (0.01ug/ml) cell lysate and human brain cerebellum(C) (0.03ug/ml), human plasma (D) (0.03ug/ml) and human testes(E) (0.03ug/ml) tissue lysates (35ug protein in RIPA buffer) were resolved on a SDS PAGE gel and blots were probed with the chimeric rabbit version of SAIC-43B-8 (NBP3-20109) before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.

Immunocytochemistry/Immunofluorescence: Clusterin/APOJ Antibody (SAIC-43B-8) - Azide and BSA Free [NBP3-20109] - Immunofluorescence analysis of paraformaldehyde fixed HeLa cells permeabilized with 0.15% Triton stained with the chimeric rabbit IgG version of SAIC-43B-8 (NBP3-20109) (1:100 dilution) for 1h followed by Alexa Fluor(R) 488 secondary antibody (1:1000 dilution), showing cytoplasmic staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP3-20109, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody followed by staining with Alexa Fluor(R) 488 secondary antibody.

Flow Cytometry: Clusterin/APOJ Antibody (SAIC-43B-8) - Azide and BSA Free [NBP3-20109] - Paraformaldehyde fixed HeLa cells permeabilized with 0.5% Triton were stained with anti-unknown specificity antibody (isotype control, black line) or the rabbit IgG version of SAIC-43B-8 (NBP3-20109, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor(R) 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

Background: Clusterin

Clusterin, also designated complement lysis inhibitor (CLI), apolipoprotein J (APOJ), sulfated glycoprotein 2 (SGP2), SP-40 and testosterone-repressed prostate message 2 (TRPM2), is a secretory, heterodimeric glycoprotein that influences immune regulation, cell adhesion, transformation, lipid transportation, tissue remodeling, membrane recycling and cell-cell interactions. Clusterin is synthesized as a 449 amino acid poly-peptide that is posttranslationally cleaved at an internal bond between Arg 227 and Ser 228. Two subunits, alpha and beta, are associated through disulfide bonds. The beta subunit (also called ApoJalpha) corresponds to residues 23-227. The alpha subunit (also called ApoJbeta) corresponds to residues 228-449. Overexpression of Clusterin appears to be more common in late stages of mammary tumor progression. Clusterin markedly influences beta-Amyloid structure and neuritic toxicity in vivo and may influence Alzheimer's disease pathogenesis.
