Application Notes
IHC: In an ABC method, we suggest an incubation period of 30 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation conditions and antibody dilutions should be determined by the user. Formalin fixed paraffin embedded tissue sections require high temperature antigen unmasking with 10 mM citrate buffer, pH 6.0 prior to immunostaining.
Please Note: Optimal dilutions of this antibody should be experimentally determined.

Background: ERCC1

ERCC1, also known as RAD10, COFS4 and UV20, is a gene that functions in the nucleotide excision repair pathway. Specifically, ERCC1 is required for the repair of DNA lesions induced by UV light or formed by electrophilic compounds. ERCC1 is a heterodimer composed of ERCC1 and XPF/ERRC4 and is located in the nucleus. Mutations in ERCC1 result in cerebrooculofacio skeletal syndrome, as well as carcinogenesis. Recent research on DNA repair suggests that DNA damage may drive accelerated bone aging (PMID: 23281008), and that defaults in DNA repair mechanisms are prevalent in many lung cancers (PMID: 23276504).
