Immunogen

This recombinant antibody was prepared by cloning the single chain fragment (scFv) derived from the murine anti-human Fc gamma RI/CD64 mAb m22 into the bacterial expression vector pBM1.1, and fusing it to a deletion mutant of Pseudomonas exotoxin A.

Specificity

This immunotoxin is specific for human Fc gamma RI/CD64, and specifically an epitope outside the Fc binding domain. The binding activity of the VH/VL antibody format was unaffected by fusion of the m22(scFv) coding regions to the truncated ETA coding sequences

Description

CoraFluor(TM) 1 is a high performance terbium-based TR-FRET (Time-Resolved Fluorescence Resonance Energy Transfer) or TRF (Time-Resolved Fluorescence) donor for high throughput assay development. CoraFluor(IM) 1 absorbs UV light at approximately 340 nm, and emits at approximately 490 nm, 545 nm, 585 nm and 620 nm. It is compatible with common acceptor dyes that absorb at the emission wavelengths of CoraFluor(TM) 1. CoraFluor(TM) 1 can be used for the development of robust and scalable TR-FRET binding assays such as target engagement, ternary complex, protein-protein interaction and protein quantification assays.

Background: Fc gamma RI/CD64

CD64 is a 72 kD single chain type I glycoprotein also known as Fc gammaRI and FcR I. CD64 is a member of the immunoglobulin superfamily and is expressed on monocytes/macrophages, dendritic cells, and activated granulocytes. The expression can be upregulated by IFN- gamma stimulation. CD64 binds IgG immune complex. It plays a role in antigen capture, phagocytosis of IgG/antigen complexes, and antibody-dependent cellular cytotoxicity (ADCC).
