Description

CoraFluor(TM) 1 is a high performance terbium-based TR-FRET (Time-Resolved Fluorescence Resonance Energy Transfer) or TRF (Time-Resolved Fluorescence) donor for high throughput assay development. CoraFluor(IM) 1 absorbs UV light at approximately 340 nm, and emits at approximately 490 nm, 545 nm, 585 nm and 620 nm. It is compatible with common acceptor dyes that absorb at the emission wavelengths of CoraFluor(TM) 1. CoraFluor(TM) 1 can be used for the development of robust and scalable TR-FRET binding assays such as target engagement, ternary complex, protein-protein interaction and protein quantification assays.

Background: GSK-3 alpha

Glycogen synthases kinase 3 alpha (GSK3-alpha) is a multifunctional Serine/Threonine protein kinase, one of two isoforms of GSK3 (1). Originally identified as a regulator of glycogen synthase, it has been shown to be involved in the hormonal control of several regulatory factors and transcription factors. GSK3-alpha activity is down regulated by phosphorylation on Serine 21 by PKB, or activated by phosphorylation on Tyrosine 279 (2). Insulin inactivates GSK3-alpha by rapid phosphorylation which leads to activation of Glycogen Synthase in the cells (3). GSK3-alpha activitiy has been linked to a pathogenic mechanism in Alzheimer's disease (4).
