Human PBMCs were treated with Recombinant Human IL-2 (202-IL,20 ng/mL) and TGF-beta (100-B,10 ng/mL) for 24 hours, then stained with (A) Rabbit anti-Human CD25/IL-2 Ra Monoclonal Antibody (Catalog # MAB10203) or (B) Rabbit IgG control antibody (MAB1050), followed by PE-conjugated Anti-Rabbit IgG Secondary Antibody (F0110) and Mouse anti-Human CD3 APC-conjugated Monoclonal Antiobdy (FAB100A). In a functional flow cytometry test, Recombinant Human IL-2 Biotinylated protein (BT202, 100 ng/mL) binds to HEK293 human embryonic kidney cell line transfected with recombinant human CD25/IL-2R alpha. (A) Binding is completely blocked by 2.5 µg/mL of Human Anti-Human CD25/IL-2R alpha Monoclonal Antibody (Catalog # MAB10203) but not by (B) Rabbit IgG Isotype Control (MAB1050) at the same concentration.
