Scientific Data Images for Human Integrin  alpha5/CD49e Antibody

Detection of Integrin alpha 5/CD49e antibody in K562 Human Cell Line antibody by Flow Cytometry.

Detection of Integrin alpha 5/CD49e in K562 Human Cell Line by Flow Cytometry.

K562 human chronic myelogenous leukemia cell line was stained with Mouse Anti-Human Integrin alpha 5/CD49e Monoclonal Antibody (Catalog # MAB1864, filled histogram) or isotype control antibody (Catalog # MAB002, open histogram) followed by APC-conjugated anti-Mouse IgG Secondary Antibody (Catalog # F0101B). Integrin  alpha5/CD49e in IMR-90 Human Cell Line.

Integrin  alpha5/CD49e was detected in immersion fixed IMR-90 human lung fibroblast cell line using Mouse Anti-Human Integrin  alpha5/CD49e Monoclonal Antibody (Catalog # MAB1864) at 8 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; NL007) and counterstained with DAPI (blue). Specific staining was localized to cell membrane and cytoplasm. Staining was performed using our protocol for Fluorescent ICC Staining of Non-adherent Cells.

Applications for Human Integrin  alpha5/CD49e Antibody

Application
Recommended Usage

Adhesion Activation

The adhesion of K562 human chronic myelogenous leukemia cells (5 x 104 cells/well) to immobilized Bovine Fibronectin (Catalog # 1030-FN, 10 µg/mL, 100 µL/well) was maximally enhanced (2-2.5 fold) by pre-treatment of cells with 500 ng/mL of the antibody.

CyTOF-ready

Ready to be labeled using established conjugation methods. No BSA or other carrier proteins that could interfere with conjugation.

Flow Cytometry

0.25 µg/106 cells
Sample: K562 human chronic myelogenous leukemia cell line

Immunocytochemistry

8-25 µg/mL
Sample: Immersion fixed IMR‑90 human lung fibroblast cell line
Please Note: Optimal dilutions of this antibody should be experimentally determined.

Background: Integrin alpha 5/CD49e

The Integrin alpha5 subunit, also called CD49e, associates with the Integrin beta1 subunit (CD29) to form the VLA-5 complex. It functions as a receptor for fibronectin and is expressed on fibroblasts, endothelial cells, epithelial cells, platelets and some melanoma cell lines (1, 2).
