Detection of Human Melanotransferrin/CD228 by Western Blot.
Western blot shows lysates of SK-Mel-28 human malignant melanoma cell line and Hs 294T human melanoma cell line. PVDF membrane was probed with 1 µg/mL of Mouse Anti-Human Melanotransferrin/CD228 Monoclonal Antibody (Catalog # MAB8175) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for Melanotransferrin/CD228 at approximately 90-95 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.Melanotransferrin/CD228 in Human Melanoma Tissue.
Melanotransferrin/CD228 was detected in perfusion fixed frozen sections of human melanoma tissue using Mouse Anti-Human Melanotransferrin/CD228 Monoclonal Antibody (Catalog # MAB8175) at 5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC001). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to plasma membrane and cytoplasm. Simple Western lane view shows lysates of SK‑Mel‑28 human malignant melanoma cell line, loaded at 0.5 mg/mL. A specific band was detected for Melanotransferrin/CD228 at approximately 109 kDa (as indicated) using 10 µg/mL of Mouse Anti-Human Melanotransferrin/CD228 Monoclonal Antibody (Catalog # MAB8175) .
