Detection of Myocilin in HEK293 cells by Flow Cytometry.

HEK293 cells transfected with Human MYOC (filled histogram) vs Irrelevant transfectant cells (open histogram) were stained with Mouse Anti-Human Myocilin Monoclonal Antibody (Catalog # MAB34461), followed by Allophycocyanin-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # F0101B). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005). Detection of Myocilin in Human Skeletal Muscle.

Myocilin was detected in immersion fixed paraffin-embedded sections of Human Skeletal Muscle using Mouse Anti-Human Myocilin Monoclonal Antibody (Catalog # MAB34461) at 15 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC001). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using VisUCyte Antigen Retrieval Reagent-Basic (Catalog # VCTS021). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to sarcoplasm in cardiomyocytes. Background: Myocilin

Myocilin (also known as TIGR) is a variably glycosylated, 65 kDa, secreted polypeptide that belongs to the family of olfactomedin-related proteins. Human Myocilin is synthesized as a 490 aa precursor that contains an 18 aa signal sequence, an 84 aa N-terminus, a 53 aa alpha‑helical leucine zipper and a 335 aa C-terminus that contains a 260 aa OLF-domain. An alternate start site generates a signal-sequenceless 504 aa mature protein. Myocilin forms nondisulfide-linked dimers and multimers. The human 32 kDa OLF-domain shares 87% aa sequence identity with the OLF-domain in bovine, rat, and mouse.
