Western Blot: IFN-gamma Antibody (D9D10) [NBP2-81044] - Western Blot using anti-IFN gamma antibody (D9D10) [NBP2-81044]. Human thymus lysate (35ug protein in RIPA buffer) was resolved on a 15% SDS PAGE gel and blots probed with the rabbit IgG-chimeric version of D9D10 [NBP2-81044] at 0.1 ug/ml before detection using an anti-rabbit secondary antibody. Protein was detected by chemiluminescence. The expected unmodified running size for IFN gamma is 19.3kDa, though the form shown here is likely glycosylated. NBP2-81044 successfully detected IFN gamma in human thymus samples.

Application Notes
This chimeric rabbit antibody was made using the variable domain sequences of the original murine IgG1 format, for improved compatibility with existing reagents, assays and techniques. This antibody can be used to detect both native and recombinant intereferon gamma (IFNg). It is reported only to bind to the functional, dimeric form of IFNg.

Background: IFN-gamma

IFN-gamma, a type II interferon and an acid-labile interferon, is mainly produced by CD4 and CD8 T-lymphocytes and natural killer cells activated by antigens, mitogens or alloantigens. IFN-gamma shows no significant homology with IFN-beta or IFN-alpha. IFN-gamma has antiviral and antiparasitic activities and exhibits also immunoregulatory and proinflammatory activities. IFN-gamma induces cytokine production and promotes antigen presentation by up regulating the expression of MHC proteins as well as other antigen proteins. In contrast to other interferons, which are mostly antiviral, the main activity of IFN-gamma seems to be immunomodulatory. Human IFN-gamma is species-specific and is only biologically active in human and primate cells.
