Western blot shows lysates of TM4 mouse sertoli cell line and mouse small intestine tissue. PVDF membrane was probed with 2 µg/mL of Rat Anti-Mouse CD155/PVR Monoclonal Antibody (Catalog # MAB6909) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). A specific band was detected for CD155/PVR at approximately 80 kDa (as indicated). This experiment was conducted under non-reducing conditions and using Immunoblot Buffer Group 1.

CD155/PVR was detected in immersion fixed adult mouse thymocytes using Rat Anti-Mouse CD155/PVR Monoclonal Antibody (Catalog # MAB6909) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Rat IgG Secondary Antibody (red; Catalog # NL013) and counterstained with DAPI (blue). Blockade of Receptor-ligand Interaction

In a functional ELISA binding assay, 0.2-0.8 μg/mL of this antibody will block 50% of the binding of Recombinant Mouse CD155/PVR Fc Chimera (Catalog # 9670-CD) to immobilized Recombinant Mouse CD96 (Catalog # 5690-CD) coated at 1 μg/mL (100 μL/well). At 10 μg/mL, this antibody will block >90% of the binding.

Background: CD155/PVR

CD155, also known as PVR (poliovirus receptor) and Necl-5 (Nectin-like molecule-5), is a 70 kDa type I transmembrane glycoprotein that binds poliovirus as well as Vitronectin, Nectin-3, and DNAM-1. CD155 contains three Ig-like domains in its extracellular region. It is expressed in a variety of normal tissues and is upregulated in tumor cells of epithelial and neuronal origin. Within aa 29-348 of the extracellular region, mouse CD155 shares 45% and 73% aa sequence identity with human and rat CD155, respectively.
