Specificity

Detects mouse IL-27 in direct ELISAs. In direct ELISAs, approximately 10% cross-reactivity with recombinant mouse (rm) IL‑27 p28, rmIL‑27 EBI3, recombinant human (rh) IL-27 p28, and rhIL-27 EBI3 is observed.

Detection of IL-27 in Mouse Splenocytes by Flow Cytometry.

Mouse splenocytes untreated (open histogram) or treated with 1 µg/mL LPS for 24 hours (filled histogram) were stained with Mouse IL-27 Monoclonal Antibody (Catalog # MAB18342) or isotype control antibody (Catalog # MAB006, not shown), followed by Allophycocyanin-conjugated Anti-Rat IgG F(ab')₂Secondary Antibody (Catalog # F0113). To facilitate intracellular staining, cells were fixed with paraformaldehyde and permeabilized with saponin.

Background: IL-27

Interleukin 27 (IL-27) is a secreted heterodimeric cytokine comprised of the IL-12 p35-related protein, p28, and the IL-12 p40-related protein, EBI3 (Epstein-Barr virus‑induced gene 3). IL-27 is expressed by monocytes, endothelial cells and dendritic cells. It binds TCCR/WSX-1 on naïve CD4⁺ T cells and induces the expression of a functional IL-12 receptor, allowing IL-12-induced Th1 polarization. Human EBI3 is 61% amino acid (aa) identical to mouse EBI3 and includes an 20 aa signal peptide and a 209 aa mature protein with two fibronectin type III domains.
