Immunocytochemistry/Immunofluorescence: NF-L Antibody [NBP1-05219] - View of mixed neuron/glial cultures stained with NBP1-05219 (red) and rabbit antibody to phosphorylated NF-H (NB300-135, green). The NF-L protein is assembled into neurofilaments which are found throughout the axons, dendrites and perikarya of these cells. In contrast the phosphorylated NF-H has a much more restricted expression pattern, being found only in developed axonal neurofilaments. Since both proteins are found in neurofilaments, the red and green patterns overlap, so that neurofilaments containing NF-L and phosphorylated NF-H appear yellowish. In contrast neurofilaments containing only NF-L appear red.

Immunohistochemistry Free-Floating: NF-L Antibody [NBP1-05219] - Staining of rat cerebellum section stained with chicken pAb to NF-L, NBP1-05219, dilution 1:2,000 in green, and costained with mouse mAb to FOX3/NeuN, dilution 1:5,000 in red. Following transcardial perfusion of rat with 4% paraformaldehyde, brain was post fixed for 24 hours, cut to 45uM, and free-floating sections were stained with above antibodies. NBP1-05219 antibody labels perikarya and processes of neuronal cells, particularly strongly the axons of basket cells, while the FOX3/NeuN antibody stains the nuclei and proximal cytoplasm of neurons.

Background: NF-L

Neuronal intermediate filaments or neurofilaments are found specifically in neurons where they play a key role in maintaining neuronal morphology as well as in regenerating myelinated axons. Neurofilaments contain three subunits: a light polypeptide (NEFL/NFL), a medium polypeptide (NEFM/NFM), and a heavy polypeptide (NEFH/NFH), with molecular weights of 68, 160, and 212 kD respectively. 68kD Neurofilament Light (NEFL) is the most abundant of three neurofilament proteins and, like other nonepithelial intermediate filament proteins, it can form homopolymeric 10-nm filaments. NEFL forms the backbone of neurofilament and perturbations in its expression leads to motor neuron diseases, such as Charcot-Marie-Tooth disease, type 2E (CMT2E). In addition to its influence on the nervous system, NEFL has been shown to act as a tumor suppressor and has been linked to carcinogenesis/metastasis of several malignancues including breast, prostate, lung, colon, and bladder cancer.
