Flow Cytometry: Notch-1 Antibody (E6) - Chimeric [NBP2-62557] - Flow-cytometry using the anti-Notch 1 antibody E6. Mouse thymocytes were stained with unimmunized rabbit IgG antibody (black line) or the rabbit-chimeric version of E6 (NBP2-62557, blue line) at a concentration of 10 ug/ml for 30 mins at RT. After washing, bound antibody was detected using anti-rabbit IgG JK (FITC-conjugate) antibody at 2 ug/ml and cells analyzed on a FACSCanto flow-cytometer.

Immunofluorescence: Notch-1 Antibody (E6) - Chimeric [NBP2-62557] - Immunofluoresence staining of fixed HeLa cells with anti-Notch 1 antibody E6 Immunofluorescence analysis of paraformaldehyde fixed HeLa cells, permeabilized with 0.15% Triton stained with the chimeric rabbit IgG version of E6 (NBP2-62557) at 10 ug/ml for 1h followed by Alexa Fluor 488 secondary antibody (1 ug/ml), showing membrane and cytoplasmic staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP2-62557, DAPI, merged channels and a negative control. The negative control was stained with unimmunized rabbit IgG followed by Alexa Fluor 488 secondary antibody.

Background: Notch-1

Notch is synthesized in the endoplasmic reticulum as an inactive form which is proteolytically cleaved by a furin-like convertase (S1 cleavage) in the trans-golgi network before it reaches the plasma membrane to yield an active, ligand-accessible form. Cleavage results in a C-terminal fragment N(TM) and a N-terminal fragment N(EC). Following ligand binding, it is cleaved (S2 cleavage) by TNF-alpha converting enzyme (TACE) to yield a membrane-associated intermediate fragment called Notch extracellular truncation (NEXT). This fragment is then cleaved by presenilin-dependent gamma-secretase (S3 cleavage) to release the intracellular domain (NICD) from the membrane which then translocates to the nucleus to activate transcription of downstream genes.
