Product Overview

The RNAscope™ Multiomic LS Assay enables highly sensitive and specific detection of both RNA and proteins in the same tissue section for a true multiomic spatial assay. Proven class-leading single-molecule RNAscope technology is leveraged to enable simultaneous detection of up to 6 total proteins and RNA targets with single cell resolution.

This assay is based on the same technology as the RNAscope Multiplex v2 assay and is compatible with your existing probes and workflows. With both protease and a new completely protease-free workflow, the kit provides significantly upgraded capabilities and benefits.

The RNAscope™ Multiomic LS Assay is designed as an automated assay for the Leica Biosystems’ BOND RX Research Advanced Staining System

Technology Overview

The RNAscope Multiomic LS assay includes two key innovations to enable the best RNA in situ hybridization results AND the best protein detection, on the same slide.

Protease-free Workflow

Expanded Protein Detection: The new RNAscope Protease-Free workflow now empowers you to use any IHC-validated antibody to co-detect proteins and RNAs on the same tissue section.

Preserved Tissue Morphology: The protease-free approach preserves tissue morphology, ensuring that your results are both precise for analysis and visually stunning.

Flexible Assay Design: Kit includes both Protease and Protease-Free reagents, so you can choose from either pretreatment options.

Ease of implementation: The Protease-Free workflow with PretreatPro seamlessly integrates with your existing RNAscope protocols.

RNA Detection

RNAscope Chemistry

Visualize RNAs with RNAscope Multiplex v2 detection technology. RNA-specific target probes using proprietary double ZZ design are amplified using RNAscope technology followed by fluorescent TSA detection.

Protein Detection

Visualize proteins using ready-to-use pre-conjugated primary antibodies or your own unconjugated primary antibodies together with available pre-qualified secondary antibodies. Antibodies are detected using RNAscope amplification technology followed by fluorescent TSA detection.

Fast & Flexible Assay Design

The RNAscope Multiomic protease-free workflow enables easy and robust integration with sequential RNA and protein detection workflows.

For increased multiomic capability, the assay can be used with primary antibodies selected from a pre-qualified panel targeting the most common cell markers. These panels ensure sensitivity and specificity and that the antibodies work together in the assay, minimizing optimization time and effort. Select up to four protein targets from the panel and combine with your own primary antibodies and available conjugated secondaries for added protein detection capability.

Finally, add probes to any gene to quickly build your own custom assay.
