CD81 was detected in immersion fixed Jurkat human acute T cell leukemia cell line (left panel; positive staining) and HepG2 human hepatocellular carcinoma cell line (right panel; negative staining) using Mouse Anti-Human CD81 Monoclonal Antibody (Catalog # MAB46151) at 8 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). CD81 was detected in immersion fixed paraffin-embedded sections of human tonsil using Mouse Anti-Human CD81 Monoclonal Antibody (Catalog # MAB46151) at 5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC001). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cell surface. Background: CD81
CD81, also known as TAPA-1 and TM4SF4, is a widely expressed protein in the tetraspanin family. CD81 is a multifunctional protein that interacts with a variety of other molecules, including tetraspanins, and is important for organization of the plasma membrane into microdomains. CD81 facilitates B cell and T cell activation and is an integrin-binding adhesion molecule. CD81 expression on lymphocytes is altered during infection by hepatitis C virus or HIV-1 and contributes to the pathogenicity of those viruses. Human CD81 shares 92%‑93% aa sequence identity with mouse and rat CD81.
