Human Ig Kappa Light Chain Antibody

Detection of IgKappa in Human Tonsil via seqIF™ staining on COMET™

IgKappa Antibody was detected in immersion fixed paraffin-embedded sections of human Tonsil using Mouse Anti-Human IgKappa, Monoclonal Antibody (Catalog # MAB100501) at 1ug/mL at 37 ° Celsius for 4 minutes. Before incubation with the primary antibody, tissue underwent an all-in-one dewaxing and antigen retrieval preprocessing using PreTreatment Module (PT Module) and Dewax and HIER Buffer H (pH 9; Epredia Catalog # TA-999-DHBH). Tissue was stained using the Alexa Fluor™ 555 Goat anti-Mouse IgG Secondary Antibody at 1:100 at 37 ° Celsius for 2 minutes. (Yellow; Lunaphore Catalog # DR555MS) and counterstained with DAPI (blue; Lunaphore Catalog # DR100). Specific staining was localized to the membrane. Protocol available in COMET™ Panel Builder.

Detection of Human Ig Kappa Light Chain by Western Blot.

Western blot shows human kappa light chain protein, human lambda light chain protein, human serum, primate serum, mouse serum, and rat serum. PVDF membrane was probed with 2 µg/mL of Mouse Anti-Human Ig Kappa Light Chain Monoclonal Antibody (Catalog # MAB100501) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for Ig Kappa Light Chain at approximately 25 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Detection of Human Ig Kappa Light Chain by Simple Western^TM.

Simple Western lane view shows lysates of human kappa light chain protein, human lambda light chain protein (negative control), and human serum, loaded at 0.2 mg/mL. A specific band was detected for Ig Kappa Light Chain at approximately 32 kDa (as indicated) using 20 µg/mL of Mouse Anti-Human Ig Kappa Light Chain Monoclonal Antibody (Catalog # MAB100501) . This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.
