Immunocytochemistry/Immunofluorescence: Numb Antibody [NB500-178] - Rat FR cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.05% Triton X-100 in PBS for 5 minutes. The cells were incubated with (NB500-178) at 1ug/ml overnight at 4C and detected with an anti-rabbit DyLight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.
Immunocytochemistry/Immunofluorescence: NUMB Antibody [NB500-178] - The NUMB antibody was tested in HepG2 cells at a 1:500 dilution against Dylight 488 (Green). Alpha-tubulin and nuclei were counterstained against Dylight 550 (Red) and DAPI (Blue), respectively.

Application Notes
In WB one may see any or all of the four isoforms. The molecular weight of human and mouse NUMB is 72 kDa (isoform 1), 66 kDa (isoform 2), 71 kDa (isoform 3), and 65 kDa (isoform 4). Multiple non-specific bands may be seen with lower dilutions and/or longer exposure times than suggested in the protocol we used to obtain our data.

In Simple Western only 10 - 15 uL of the recommended dilution is used per data point. Separated by Size-Wes, Sally Sue/Peggy Sue.
Please Note: Optimal dilutions of this antibody should be experimentally determined.

Background: Numb

Numb is a transmembrane signaling receptor that is activated by the DSL family of ligands. Studies suggestthat mammalian Numb plays a role in progenitor cell fate in neurogenesis. Numb exerts its role on cell fate by antagonizing Notch signaling. The mammalian Numb gene undergoes alternative splicing to produce at least four functionally different Numb isoforms which regulate different processes. A loss of Numb expression has been demonstrated in breast cancer, non-small cell lung cancer and chronic myelogenous leukemia.
