Background: Numb

Numb is an evolutionary conserved protein which plays a crucial role in cell fate determination, an important event in embryonic development as well as in certain pathologies including cancer. Numb was initially identified in Drosophila (dNumb) as a membrane-associated protein which asymmetrically segregates in dividing cells and define distinct cell fates by interacting with/inhibiting Notch. dNumb's mammalian homologs (encoded by two genes mNumb and mNumb-like) displays a higher degree of complexity with alternative spliced isoforms, and display multitude of functions such as control of asymmetric cell division and cell fate choice, endocytosis, cell adhesion, cell migration, ubiquitination of specific substrates and regulation of Notch, Hedgehog, p53 etc. The multiple functions of Numb highlight its critical role not only in the control of physiological processes, but also in diseases, when deregulated. The ability of Numb to control oncogenic (Notch and Hedgehog) and tumor suppressor (p53) pathways provides a strong link between deregulated Numb function and cancer. Indeed, hyperactive Notch and Hedgehog functions have been described to be responsible of several tumors including breast cancer and medulloblastoma.
